Wound healing in an infectious environment

Pseudomonas aeruginosa (PA) biofilm plays an undisputable role in the local microenvironment of chronic wounds. The main focus of our research is the inflammatory response to PA biofilm in the skin and to evaluate a potential clinical implication of PA biofilm infection on skin regeneration capability. PA biofilm suppresses local host response. The usefulness of an innate host defense antimicrobial peptide in infected murine wounds is currently evaluated.

We use two immunologically diverse genetically inbred mouse strains, C3H/HeN and BALB/C. Based upon previous findings that PA biofilm infection aggravates local skin inflammation evaluated by IL-1β in our chronic wound model, we aim to further elucidate the role of host response towards PA biofilm wound infections in vivo. Full-thickness burn wound inflammatory innate host response towards PA biofilm infection is evaluated eg. at day 14 post wounding.  Subcutaneous injection of 100 μl of a PA biofilm solution (107 CFU/ml) is carried out at day 4 post wounding in order to circumvent burn wound induced systemic immunosuppression.
