The Ph.d.-project Amyotrophic Lateral Sclerosis(ALS) and the Innate Immune System are establishing a national ALS biobank. The goal of the project is to study the innate immune response in ALS patients with focus on the complement system.

The hypotheses of study:

1. ALS is characterized by an abnormal immune response with cytotoxicity and increased complement activity in plasma which is revealed by the following:

a. plasma from patients shows increased haemolytic activity when incubated with red blood cells from healthy subjects.

b. the complement activity is increased in the plasma from ALS patients compared with plasma from neurologically healthy subjects and subjects with other neurological disease.

c. The haemolytic activity in the plasma from ALS patients is correlated with the complement activity.

2. Spinal liquid from ALS patients contains increased complemtn activity compared with spinal liquid from neurologically healthy subjects and from subjects with other neurological disease.

The project consists of 4 clinical studies:

1: Haemolytic activity and the complement system in ALS plasma

2. Case-control study aiming to mapping the complement system

3. The complement system of ALS patients over time -a cohort study

4. Searching for complement activity in the neuromuscular junctions
